I just posted a community question at the Twitch Support Center:
http://help.twitch.tv/customer/portal/questions/9868519-video-highlights-unplayable (http://help.twitch.tv/customer/portal/questions/9868519-video-highlights-unplayable)
I hope that someone is actively investigating the problem.
Disheartening news, if the affected video highlights are permanently inaccessible. :(

Hey guys,
Check the listing of archived highlights on your own Twitch channels.
You may not like what you see.

Huh... that's strange and rather scary..
I just checked a couple of my highlights, they seem okay but will check others when time allows. If it's a video that I really value, then I usually export to Youtube also, and sometimes also download the video from Youtube, so it exists in multiple places.
I hope this gets worked out BibleThump FailFish

The problem seems to be most prevalent with clips that were created prior to 2013.

The problematic videos are easy to spot. Notice the blank areas where a screen capture should appear:

This was happening to me during last DKO. Looks exactly the same except at least one of the videos would say 10hrs long even though I only played four.

I've been submitting requests on some of my videos for months, as they only show a black screen and don't play. I did notice this problem for the first time last night as I was doing my (probably bi-monthly) rounds requesting they fix my videos.
